Can someone who is an Indian Passport holder with valid multiple entry visas to Canada and USA come to Toronto first and then enter US and return back to Canada? Are any specific forms to be filled before entering US or is the multiple entry visa enough?

Yes.
When you enter US for the first time for the trip, they will process the US-VISIT (fingerprinting and photographing) and issue a multiple entry I-94 that will be valid for upto 6 months.
That's it. After that, you can enter both countries any number of times within the Canada visa validity and within I-94 validity. The last time you leave US, you may want to surrender the I-94 so that your final "departure" is duly official.
The above is for land crossings.
In the case of flying between US and Canada, they may issue a new I-94 each time you want to enter US.
