It's actually quite a reasonable drive from Mississauga to Guelph in the winter (and assuming you're not in a rush, a beautiful one in the summer / autumn if you stay off the 401 and take the country roads).
During the Dec - March period the 401 is reasonably clear and on most winter days you'll take no longer than you would ordinarily any other time of the year. The only trouble spot tends to be the standstill of traffic prior to the Mavis exit in the evenings, in which case I would advise taking Mississauga Rd exit, and using Derry to get to Mavis.

The only alternative route is Highway 7 which runs parallel to 401. You have to take some diversions on Highway 7 (road is not a straight one). There are some other parallel roads north of 401 (like side road 1 or 2) but they run for short distances and you might get lost. Perpendicularly is the road which runs through the Mowahawk race track.
Even if you are in slowdown (unless a complete shutdown), I would advise you not to take either Highway 7 or the Perpendicular road through Mowahawk race course. It is pretty desolate and actually increases your traffic time significantly. Once I had a donut tire and I took hwy-7 from islington to Guelph and it took me 2 hrs. to reach guelph (at 8:00 pm).
In snowy days, just forget that route. It will be a night mare.

Hwy 7 is awesome only if you live in Brampton or the North West end of Mississauga.
If you are in any other area then its only 401

You will find that commuting to guelph on a normal day for 50 minutes is less stressfull than driving for 25 mins in peak mississauga traffic
In Guelph there is only rush minute (no rush hour) :-)
If you dont know already, when you are driving from Mississauga take the Hwy 6 S exit instead of the Hwy 6 N exit and go north. (it is one ext before)
Then drive along gordon st (also called brock road) till you reach stone road, you may save 5 mins (depending on traffic) but you will definately save 2$ worth of gas in one day
